The A6 starts at a roundabout with the A505 as St Mary's Street and Guildford Street in Luton. From here, it forms a de facto ring road to the north of the town centre, before meeting its original course north. There's a brief dual carriageway bypass of Barton-le-Clay and further dual sections up to Bedford. The road meets the A421 bypass at a grade-separated roundabout junction, the road ahead being the A5141.
Since 2013, the A6 has headed around Bedford via the A421 and A428 bypasses, ceding its number to the former but asserting itself over the latter. At Bromham it turns east (where the A428 turns off west to Northampton) and briefly runs along the former A4280 to Biddenham. Since 2016 a newly-built stretch of road, the Great Ouse Way, carries the A6 north and then east over the Midland Main Line to a roundabout south of Clapham, where it meets the A600 coming up along its original route out of Bedford town centre.
