The C288 is a rather lengthy route which starts on the A2 near Ballykinler. The road heads northwards inland and eventually meets the A25 between Downpatrick and Clough. North of the A25, the road crosses the C289 at Magheralagan Lake. The road crosses the C278 via a short multiplex next to Loughinisland Lake. From here the road continues north west until it reaches a mini roundabout on the edge of the village of Drumaness. The C288 does not continue into the village however, but the curiously numbered C388 travels the rest of the way through the village to the A24.
