The C28 starts on the B955 at Dykehead, and climbs between the wooded heights of Mile Hill to the south and Tulloch Hill, with the Airlie Memorial Tower on its summit, to the north. At a right-hand bend where the Scott-Wilson Memorial Sculpture is situated, the road turns north into Glen Prosen.
Following the east side of the glen, at first the road remains a long way above the river, but then starts to drop to the bridge over the Glencally Burn near Buckhood, before turning southward for a few yards and then resuming a north-westerly course again as it climbs, still above the Prosen Water.
It then drops down to the riverbank over the next couple of miles to reach the bridge at Spott. Here, a short spur (the C28-1) crosses the bridge to meet the C27, whilst the main road continues north to Glenprosen Village.
