The C307 is a very long route in County Down. Starting on the A50 at Katesbridge, the road heads north east. The road crosses the C322 before coming to a pair of sharp bends which results in the road briefly deviating from its north eastward travel to head north west. To the north of here, the road passes through a crossroads at Enagh with the C310 to the left and the C308 to the right. After crossing the Tullynisky Cut, the road passes through the village of Kinallen where it crosses the C328. The road heads north east as it crosses the upper portion of the River Lagan before passing the C327. The road crosses the B2 and the C306 before passing by Lough Aghery. In Annahilt, the road crosses the B177 at a crossroads. The road ends on the C297 at Glebe after over 20 kilometres.
