The C401 heads east from the A79 in the centre of Prestwick. From The Cross, it follows Alexandra Avenue, and then zigzags along Caerlaverock Road into Brerelands Road. Continuing east, this becomes Sherwood Road, before the C401 comes to an end in the industrial area on the edge of the airport. The route previously continued east to meet the A719 (now the line of the A77 bypass) before the airport was extended.
