The C421 is a T shaped route in east Fermanagh. It starts in the village of Clabby at a mini roundabout on the B107 and heads west towards the B80. The road crosses Many Burns River before coming to a T junction where the main route TOTSOs to the south, with the spur to the west leading towards the B80. The road to the south crosses over Many Burns River again before meeting the B107 at a crossroads. Continuing south, the road eventually meets the C422 at a scattered crossroads. To the south the road crosses Colebrooke River before coming to an end on the C419 at Ashbrooke.
