The C45 starts on the A91 at Dairsie and heads south east, turning south, to pass under the railway line and reach a T junction. Ahead, a spur returns to the A91 further south, whilst the main line turns left to cross the River Eden. The river is then followed upstream the short distance to its confluence with the Ceres Burn, which is, in turn, followed south to Pitscottie. Here, the C45 ends at a fork on the B940 just north of the crossroads with the B939.
