The C501 starts on the B193 in Killea, a village straddling the border between County Donegal and County Londonderry. Very soon the road TOTSOs right and follows Killea Road to the north east, roughly running in parallel with the border. The road splits where it crosses Creevagh Road with the eastern segment coming to an end on Creevagh Road and the western segment continuing beyond. As the border moves away the road TOTSOs left to maintain a north easterly direction of travel, soon turning east at another T junction. The road ends as it meets the B527 Creggan Road in Derry, entering the city as it TOTSOs onto Hollyhall Road.
