The road starts on the A44 (former B63) at The Drones and heads westward. After going through Killyramer the B15 is met. The road then crosses the Belfast to Derry railway line at a level crossing before reaching the suburbs of Ballymoney. The A26 bypass is crossed at a roundabout and the road heads into town along Kilraughts Road to meet the B62 (pre-bypass A26) at another roundabout. The town centre is to the right.
Originally the B16 ended here; however, by 1930 it had been extended ahead. The road therefore now leaves Ballymoney to the southeast, running parallel to the A26 but with few links between the two roads. After going through Dunloy the road ends on the B93, where that road has to TOTSO. Glarryford is a short distance ahead.
