The C53 starts at the eastern end of the A80 spur at Mollinsburn Interchange and heads east through the village, before turning south east onto Mollinsburn Road. The route then doglegs to the west at Annathill, before wiggling through a crooked crossroads at South Medrox. At Greenfoot the road passes under the railway, and then steadily turns east to meet the B802 at Brackenhirst north of Glenmavis.
