Leaving the A386/A3064 junction at Milehouse in Plymouth this S2 road forms a semi-loop round the perimeter of Devonport Naval Base to meet the A3064 again.
The road heads away from the from the large A386/A3064 junction at Milehouse on its way up to Stoke Village where it twists and turns through the shopping area before TOTSOing at a mini roundabout to run down towards Morice Town. There is a short spur here that joins the A374 on its approach to the Torpoint Ferry. The B3396 continues onwards following the wall of the Naval Base connecting its various entrances and HMS Drake through Keyham before turning away from the river at Weston Mill creek to join the A3064.
Originally Milehouse Road was part of the A3041, Albert Road the A3043 and the remainder the A3042. The B3396 came into existence in the 1970s following the opening of the present route of the A38.
