The C110 starts at a crossroads on the A71, opposite the B7019, at Shawsburn and heads south east along Ashgillhead Road and Whinknowe through Ashgill. It then follows Netherburn Road to the village of the same name, but with the main road through the village closed up, the C110 takes a detour to the east around Provost Crescent. Netherburn is left on Draffan Road which continues south to Draffan and then winds south west over the M74 to meet the B7078 just north of Blackwood.
