The C126 starts at a sharp bend on the B755 in Hamilton and heads west, following Wellhall Road, Earnock Road and Newhousemill Road out of the town. The route then runs south west, still as Newhousemill Road, crossing the Rotten Calder as it reaches East Kilbride and coming to an end on High Common Road, which is not apparently a C road, just short of its junction with the A726.
