The B6100 is a short urban B-road to the south of Barnsley.
The road starts on the A61 in Worsbrough, just to the south of the Pegasus crossing, and road heads east, with the Worsbrough Country Park on its right until the road crosses the Transpennine Trail.
The B6100 then continues through Worsbrough Dale, after which there is a short semi-rural section. Now heading north, the road goes along Hunningley Lane. Just after going under the Barnsley to Sheffield railway line Hunningley Lane bears off to the left (although it is not accessible to traffic); this is the original line of the B6100, which headed straight to the A635. Now, however, the B6100 continues along a new link road to end on a roundabout where it meets the A635 and A633.
