The B6280 consists of two related B-roads either side of Darlington town centre.
The road starts at a roundabout on the B6279 in the Cockerton area of town and heads south along the residential Carmel Road, soon reaching a roundabout on the A67. Originally the B6280 continued straight across to end on the A1 (now A167) in Blackwell. However, with the construction of the Darlington bypass the B6280 and A67 swapped places at this roundabout so that the A-road could avoid the town centre. As such the B6280 now turns east at the roundabout to run along Coniscliffe Road (the original line of the A67) to meet the A167 town centre relief road at a roundabout.
There is then a short multiplex along the relief road to another roundabout, where the B6280 regains its number and heads eastwards along Parkgate along what continues to be the pre-bypass route of the A67 (although this road was numbered B1273 until the 1960s). We go under the East Coast Main Line adjacent to the station before running through the eastern suburbs of town. After going through an industrial estate the road ends at a roundabout on the A66 eastern bypass, with the A67 running ahead.
