The C72 starts in the large town of Larne, but it is not completely clear exactly where it starts. It is presumed to commence on the A2, following Main Street westwards to a mini roundabout and then heading north along Agnew Street. After the next crossroads, the C72 continues for definite along Old Glenarm Road going north. The road also includes a spur along Branch Road leading back to the A2 at the northern edge of the town. The shape of the coastline naturally forces the road to follow more of a SE/NW alignment from here as it approaches a scattered crossroads just outside Ballygally. The road continues on the other side of the crossroads and terminates on the C71 in Carncastle just a short distance north west.
