The C56 starts at a TOTSO crossroads on the B96 between Portglenone and Cullybackey. The road heads east from the crossroads and soon crosses the B93 at a junction. The road bends round to the south east and turns left onto a bridge crossing the River Maine. On the opposite side of the bridge the road includes a spur to the south past Galgorm Industrial Estate that reaches a roundabout on the A42. North from the bridge, the road continues towards the village of Cullybackey, where it meets the B62 at a T junction. Just across the road from here, the C56 continues north east over a level crossing adjacent to the village's railway station on the Belfast - Derry line. Once out of Cullybackey, the road continues a little further to meet the C57 at a crossroads. The road ahead leads to the A26 and is U-class.
