The C216 starts on the A27 at Jerrettspass opposite the C229 and after crossing the Belfast - Dublin line it TOTSOs to the south. A spur leads south west and links to the A28 from just south of here. The main line of the route continues south running alongside the A27 until a T junction is reached head-on. The road east links back to the A27 just north of the A1 Newry bypass while the road to the west leads to the A28. The road resumes its southward journey from a bit closer to the A28's interchange with the A1 and crosses the B133 into Bessbrook along Hillcrest Heights. At the end of this road, the route travels the relatively short distance along Derrymore Road to finish on the B112.
